Cinétiques de décontamination en milieu contrôlé de moules toxiques (Diarrheic Shellfish Poison). Premiers résultats ArchiMer

Laboratory assays have been performed, using mussels contamined with Dinophysis, in order to assess potential depurations kinetics. Mussels samples from Seine bay, Douarnenez bay and Vilaine bay have been tested . From these assays, which are to be performed in 1989; two main results are stressed : 1) when the toxicity level is near 0,5 MU/g of hepatopancreas*, depuration is undedectable during twelve days, 2) when the toxicity level exceeds 1 MU/g,depuration is important during the first eight days. *(quarantine level used by Japanese authorities) .

Continuous fluorescence recording as a way to improve Pacific oyster (Crassostrea gigas) models of paralytic shellfish toxin accumulation ArchiMer

A simple system was used to simulate the effect of alternating toxic (paralytic shellfish poisoning toxins) and non-toxic microalgal diets on oyster feeding behaviors and rates of toxin accumulation. These experimental conditions were meant to reflect, to some extent, the incoming and outgoing fluxes of toxic algae observed at the mouth of the Penze estuary (Northern Brittany, France). Physiological and toxicological parameters were estimated based oil fluorescence measurements recorded continuously at the outlet of each experimental tank, which contained a single oyster. Décontamination expérimentale de bivalves toxiques : application aux phycotoxines paralysantes. Rapport final ArchiMer

A sea water recirculating flume was especially designed in the framework of this study. It improves physiological and ecophysiological evaluation of a 25 oysters (c. gigas) set fed on PSP toxic A. minutum cultures and then detoxified with commonly used aquaculture species (I. galbana, T. suecica, S. costatum, T. weissflogii). Chlorophyll a levels are kept steady in each case and adjusted in order to reach the same food value in each tank (0,5 mg/l TPM). When exposed to the mildly toxic species A. minutum (0,5 pg.eq.STX.cel-1) oysters depict a reduced clearance rate, food uptake and shell-valves activity (compared to non-toxic algal diets). Ces bioessais sont : - un test sur micro crustacé dont la méthodologie est normalisée et qui est très utilisé en écotoxicologie aquatique ; - un test de cytotoxicité à court terme qui a été mis au point pour cette étude.

PSP toxin levels in cultivated mussels and oysters were monitored over a one year period (1994) in two keys areas, the Morlaix and la Penzé estuaries (Northern Brittany, France).Toxins were detected by IP-HPLC and results compared to those given by the bioassay routinely used by IFREMER Coastal Laboratories. Shellfish contamination seems closely related to A. minutum occurences (the minimum concentration of cells resulting in toxin detection in mussels was 400 cells/I). Toxins were depurated rapidly initially, and subsequently at a slower rate, such that residual toxin levels were detected in mussel digestive glands three months later. Etude de la décontamination de moules toxiques (toxines diarrhéiques) en laboratoire et en milieu naturel ArchiMer

Two studies of depuration, one in laboratory conditions and the other one in an oyster's pond were performed using 2 sets of contaminated mussels with diarrhetic toxins at a high and a low level (3 and 1 M.U.). Depuration kinetics were followed both by the adult mouse test and okadaïc acid analysis. Results are - in the pond, high toxic mussels need about 20 days to become eatable (toxicity level : 0.5 M.U.g-1 digestive glands), -this time decreases when the initial toxicity is low but it increases when mussels are underfed, - a good correlation is observed between the total toxicity and okadaïc acid concentration.

Shellfish production, and production of marine bivalve molluscs in particular, is the main form of marine aquaculture in the European Union, with a production estimated at 1,200,000 tons in 1999 (Food and Agricultural Organization). Moreover, aquaculture production is rapidly expanding throughout Europe and is becoming a major economic resource in developing areas such as Northern Greece, Ireland, and northwestern Spain. These shellfish, mainly mussels, oysters, and clams, are produced in a natural environment. L'objectif général de l'étude est de simuler les épisodes de contamination de certains bivalves (huîtres et moules) par les toxines d'Alexandrium minutum et A. calenella à partir d'un module expérimental adapté, et en vue de construire un modèle de contamination applicable à ces espèces. Ce modèle sera ensuite susceptible d'être couplé aux modèles biologiques locaux (estuaire de Penzé et étang de Thau) de déterminisme des blooms, ainsi qu'aux modèles hydrodynamiques développés par différentes équipes (Ifremer, Université de Montpellier). La première année (2001-2002) était consacrée à la simulation des contaminations en estuaire de Penzé (Alexandrium minulum) et au développement de cultures en masse d'A. calenella de Thau en bioréacteurs.

Paralytic shellfish poison accumulation yields and feeding time activity in the Pacific oyster (Crassostrea gigas) and king scallop (Pecten maximus) ArchiMer

Pacific oysters and king scallops placed individually in a recirculating flume were fed for 2 weeks with a constant concentration (120 cell ml(-1)) of a toxic strain of Alexandrium minutum. Fluorescence at the outlet of each experimental unit was measured continuously, and biodeposits were recovered twice daily to evaluate feeding time activity (FTA) and rates of organic filtration (OFR), ingestion (OIR) and organic absorption (OAR). Ion-pairing high performance liquid chromatography (IP-HPLC) was performed concurrently to quantify paralytic phycotoxins both (i) individually in shellfish at the end of the contamination period and (ii) in A. minutum cultures to estimate cellular toxin concentration. Paralytic shellfish poison outbreaks in the Penze estuary: Environmental factors affecting toxin uptake in the oyster, Crassostrea gigas ArchiMer

Several experiments using a self-regulated system were conducted to define the factors likely to influence the uptake of paralytic shellfish poison (PSP) by oysters in the Penze estuary (France, Brittany). Each 4-day experiment was carried out in a recirculated sea water system using 15 Pacific oysters (Crassostrea gigas) separated from each other and supplied with unfiltered natural seawater containing alternatively toxic (Alexandrium minutum) or non-toxic (Skeletonema costatum) algal diets.
